摘要:目的利用電噴霧離子化為接口(ESI)的液相色譜/質(zhì)譜聯(lián)用技術(shù)(HPLC—MS),鑒定重組大腸桿菌不耐熱腸毒素B亞單位(rLTB)氨基酸序列結(jié)構(gòu)的正確性。方法利用SDS—PAGE分離、呈現(xiàn)蛋白混合物,胰蛋白酶原位水解rLTB.HPLC—MS聯(lián)機(jī)制備肽指紋圖,分析肽指紋圖獲得rLTB氨基酸全序列信息。結(jié)果 rLTB的全序列結(jié)構(gòu)與理論的全序列結(jié)構(gòu)*一致。結(jié)論改進(jìn)的HPLC—MS測(cè)定條件穩(wěn)定,提高了質(zhì)譜靈敏度,結(jié)合肽指紋圖的分析結(jié)果,可以驗(yàn)證蛋白質(zhì)的氨基酸序列。 關(guān)鍵詞:rLTB;原位酶解;HPLC—MS;肽;指紋圖譜 The Structure Identification of rLTB by HPLC-MS YANG Jun,TIANG Wen—Biao,ZHU Yong—Hong,ZOU Quan—Ming (Department of Clinical Microbiology,3bird Military Medical University,Chongqing 400038 China)
ABSTRACT:Aim 3he correcTESS of the primary structure of recombinant heat—labile enterotoxin B was identified by liquid chromatography/electrospray ionization mass spectrometry.Methods rLTB in SDS— PAGE gel was digested by trypsin,an d its peptide finger mapping Was m pared by improved HPLC—MS methods.By an alyzing peptide finger mapping,the primary structure of rLTB Was identified .Results: 3heoretic an d actual primary structure of rLTB is consistent.Conclusion Improved HPLC — MS conditions were stable.3he sensitivify of MS Was increased .Whole amino acid sequence of proteins were detected .
